MagnetofectamineTM combines the renowned LipofectamineTM 2000 transfection reagent from InvitrogenTM with the unique MagnetofectionTM based reagent CombiMagTM from OZ BiosciencesTM

The combination of these two techniques in a single kit provides increased transfection efficiency, minimized toxicity and enhanced gene expression. It is particularly suited to hard-to transfect and primary cells.

 

MagnetofectionTM is a creative, simple and highly efficient method to transfect cells in vitro and in vivo. This magnetic assisted transfection technique is based on the association of magnetic nanoparticles with nucleic acids (alone or pre-complexed with a transfection reagent) to which a magnetic field is exerted. The magnetic force drives the DNA vector towards the target cells, thus causing the vector dose to get concentrated onto the cell very rapidly and trigger delivery via endocytosis. Consequently, high transfection efficiencies are achieved with a small amount of nucleic acid.

LipofectamineTM 2000 transfection reagent is a proprietary formulation of InvitrogenTM for transfecting nucleic acids into eukaryotic cells.

NOTE: As for all MagnetofectionTM applications, MagnetofectamineTM requires an appropriate magnetic field. Two magnetic plates especially designed for MagnetofectionTM are provided to exert this specific magnetic field. The special geometry of one magnetic plate produces a strong magnetic field that is suitable for 96-well plates (cat. # MF10096) and the other one (cat. # MF10000) is suitable for all culture dishes.

NOTE: This Magnetofectamine Starter Kit includes one MF10000 Universal Magnetofection Magnetic Plate; if you already have a Magnetofection Magnet, you can order the MagnetofectamineTM Transfection Kit  MTX0750 without the magnet.

RESULTS

Superior DNA transfection efficiency in primary cells

Superior transgene expression in cell lines

Classical and hard to transfect cell lines were transfected using 0.25 µg and 0.5µg pVectOZ-GFP plasmid respectively complexed with Lipofectamine™ 2000 (grey bars) or with Lipofectamine™ 2000 and CombiMag at a DNA/CombiMag ratio of 1:1 (green bars). Results were analysed by cytofluorometry 24 h after transfection.

Using Magnetofectamine™ transfection efficiencies exceeding 50% can be achieved into many primary cells, such as HUVEC, Chondrocytes and human Mesenchymal Stem Cells.

Primary cells were transfected with Magnetofectamine™ kit in 24-well plate and efficiency
was monitored 48 hours post-transfection by flow cytometry.
 

Many other classical and hard-to-transfect cell lines, such as SH-SY5Y, PC-12,
MEF, HMEC-1, C6 are transfected with high levels of transfection efficiency.

Minimize cytotoxicity

Magnetofectamine™ performs even with low doses of nucleic acids
resulting in minimized cytotoxicity. No need to increase the amount of plasmid DNA and reagent to increase transfection efficiency.
