EnPresso Y Defined Tablet medium is a ready-to-use, pre-sterilized culture medium designed for yeast cultures in small-scale cultivations from shake flask to multiwell plates. The principle of the EnPresso Tablet culture is based on the patented EnBase Technology, enzyme-based substrate delivery, which uses an enzyme to release glucose from a solubilized polysaccharide within the medium in a slow and and consistent manner. The enzymatic control of glucose delivery provides an optimal cultivation environment for yeast to reach greater cell and protein product yields.
Kit Components:
- EnPresso Y Defined Medium Tablets (orange bag): each bag contains three tablets, which consist of defined media components and a polysaccharide complex as substrate
- EnZ I'm (3,000 U/L): mixture of filter-sterilized enzymes
Each cultivation of 50mL requires one orange bag.
Storage: Store tablets at room temperature. The EnZ I'm is stable at room temperature, but should be stored at 4-8oC for long term storage. Expiration date is indicated on the package.
Additional items needed:
- Sterile shake flask*
- Sterile water
- Inducing agent (e.g. methanol)
- Antifoaming agent (eg AntiFoam 204)
- AirOtop Enhanced Seals*
Recommended Conditions:
- Volume: 10% of the flask volume
- Shaking: 250rpm, 25-50mm amplitude
- Temperature: 28-30oC is recommended
The cultivation conditions can be easily optimized for each cultivation vessel. We recommend using bromthymol blue to indicate when pH is above 7.6
Ultra Yield Flasks and AirOtop Enhanced Seals are recommended for appropriate oxygen transfer.
